Company to present data on automated solution at Association of Molecular Pathology Annual Meeting and Corporate Workshop Oct. 31
ITHACA, N.Y. — Rheonix, Inc. announces the launch of its NGS OnePrep™ solution, an integrated, automated method for DNA extraction and next-generation sequencing (NGS) library preparation on the Encompass Optimum™ workstation, at the Association of Molecular Pathology (AMP) Annual Meeting and Expo Nov.1-3 in San Antonio, Texas. The method will allow laboratory technicians to produce sequence-ready libraries directly from raw samples with significantly reduced hands-on time compared with current methods.
The NGS OnePrep solution streamlines the separate and distinct processes of nucleic acid extraction and NGS library preparation on a single instrument. Existing methods are typically highly manual or require multiple instruments to automate DNA extraction and library preparation. Even “automated” methods for library preparation have disjointed, manual steps. Utilizing the Encompass Optimum workstation, sequence-ready libraries can be produced directly from the sample in just four and a half hours and yield NGS sequence data and quality metrics indistinguishable from the more labor-intensive methods often employed by laboratory facilities. The NGS OnePrep solution offers the following benefits:
- Significantly reduced hands-on time and total time-to-results
- Ability to process up to 24 samples in four and a half hours
- Reduced intra- and inter-batch variability of NGS libraries
- Reduced lab footprint and capital equipment cost
Rheonix will host an AMP Corporate Workshop, titled “From Raw Sample to Sequence-Ready Library: Fully Integrated and Automated Nucleic Acid Extraction and NGS Library Prep System,” on Oct. 31 from 9-9:50 a.m. in room 214B of the Henry B. Gonzalez Convention Center, where collaborators will present data generated with the Rheonix solution. In both presentations, sequence-ready libraries were prepared using the Rheonix NGS OnePrep automated DNA extraction and library preparation solution. Vijaya Murugaiyan, Ph.D., research scientist at the University at Buffalo’s Center of Excellence, will describe the sequencing of DNA obtained from human saliva. The DNA libraries were prepared using Illumina Nextera DNA Flex Library Prep Kit reagents. Both human and microbiome DNA were sequenced with excellent quality metrics and alignments against the reference genomes. Patrick Martin, Ph.D., associate director at Takara Bio will describe the sequencing of DNA libraries prepared from formalin-fixed paraffin embedded (FFPE) tissue blocks, obtained from AccuRef Diagnostics, using the Takara Bio SMARTer® ThruPlex® kit reagents.
Rheonix currently offers compatibility with the Illumina Nextera DNA Flex Library Prep Kit, with plans to automate other Illumina prep kits as well as those of other manufacturers, including the Takara Bio SMARTer ThruPlex, in the near future. Initially, the NGS OnePrep solution will be compatible with the following sample types: buccal swab, saliva, fresh frozen tissue, whole blood and bacterial isolates. The solution will be expanded to include FFPE tissue and circulating cell-free DNA (ccfDNA) in the coming months.
“The ability to automatically isolate DNA and prepare sequence-ready DNA libraries with a single instrument will help to reduce the overall cost of NGS for both research and clinical applications,” said Richard Montagna, Ph.D., FACB, senior vice president for scientific and clinical affairs at Rheonix. “By integrating all DNA isolation and library preparation steps on a single workstation, the Rheonix method not only reduces a one- to two-day process to less than five hours, it also eliminates the need for multiple pieces of equipment and dramatically reduces the technical skill required to successfully isolate DNA and prepare sequence-ready DNA libraries.”
Rheonix will showcase the NGS OnePrep solution for use on the Encompass Optimum workstation at AMP booth 1525.
